目的 探討谷氨酰胺對(duì)醋酸損傷的大鼠畸形腺窩灶的影響及其機(jī)理。
方法 Wistar大鼠30只均分成對(duì)照組、醋酸組和谷氨酰胺組3組。 用1%醋酸灌注大鼠結(jié)腸制成大鼠結(jié)腸損傷模型,谷氨酰胺組在制模后第2 d 開始給予谷氨酰胺灌胃。 各組于制模14 d后處死動(dòng)物,取損傷結(jié)腸組織進(jìn)行HE染色,觀察病理組織學(xué)變化; 對(duì)所取的結(jié)腸隱窩用鹽酸浸潤(rùn)消化法分離,于掃描電鏡下觀察隱窩情況; 用免疫組織化學(xué)方法檢測(cè)結(jié)腸黏膜上皮細(xì)胞中CD44和細(xì)胞間黏附分子-1(intercellular adhesion molecule-1, ICAM-1)的表達(dá)情況。
結(jié)果 制模后14 d, 醋酸組畸形腺窩灶數(shù)目較谷氨酰胺組和對(duì)照組多,且呈分叉狀。 谷氨酰胺組結(jié)腸黏膜上皮細(xì)胞中ICAM-1和CD44表達(dá)陽性細(xì)胞數(shù)(302.1±30.1、 298.6±28.3)均明顯高于對(duì)照組(223.6±23.5、 221.5±28.6)和醋酸組(198.5±19.5、 215.3±17.8),P<0.05; 而后2組間差異無統(tǒng)計(jì)學(xué)意義(P>0.05)。
結(jié)論 谷氨酰胺能減少醋酸損傷大鼠畸形腺窩灶的形成; 增加CD44和ICAM-1的表達(dá),可能是谷氨酰胺能夠減少其形成的一個(gè)重要因素。
