目的 通過WST-8和膠原凝膠微滴腫瘤藥物敏感性檢測(CD-DST) 2種藥敏檢測方法的比較,對CD-DST法進(jìn)行評價(jià)。方法 以胰腺癌細(xì)胞系SW1990、PCT-3和ASPC-1為對象,分別采用WST-8和CD-DST法測定其對5-氟尿嘧啶(5-FU)、健擇(GEM)和奧沙利鉑(OXA)3種藥物的敏感性。結(jié)果 在一定的活細(xì)胞數(shù)目范圍內(nèi)(500~10 000個(gè)),CD-DST法所測得的細(xì)胞積分光密度與細(xì)胞數(shù)目成線性正相關(guān)(r=0.991 1, P<0.05); 3種腫瘤細(xì)胞的生長抑制率CD-DST法均高于WST-8法(P<0.05),2種方法測定的5-FU、GEM和OXA藥物敏感性結(jié)果一致。結(jié)論 CD-DST法可用于胰腺癌的體外藥敏檢測,具有一定的臨床應(yīng)用價(jià)值。
