目的探討腫瘤相關糖蛋白72(TAG72)靶向性嵌合錨定T淋巴細胞的制備方法,并檢測它對TAG72陽性肝癌細胞增殖的阻滯效應。方法分離外周血單核細胞(PBMC),然后用免疫磁珠法分離得到CD8+ T淋巴細胞。將重組真核表達載體anti-TAG72-scFv-CD28-pcDNA3.0采用脂質體介導的細胞轉染和細胞培養(yǎng),以制備TAG72靶向性的嵌合錨定T淋巴細胞; 將嵌合錨定T淋巴細胞與TAG72陽性肝癌細胞SMMC7721共培養(yǎng),通過流式細胞儀檢測肝癌細胞的周期變化,分析嵌合錨定T淋巴細胞對肝癌細胞增殖的抑制效應。結果TAG72靶向性嵌合錨定T淋巴細胞可識別肝癌細胞SMMC7721; 用流式細胞儀檢測發(fā)現,嵌合錨定T淋巴細胞可引起肝癌細胞SMMC7721的增殖阻滯。結論TAG72靶向性嵌合錨定T淋巴細胞可特異性識別TAG72陽性肝癌細胞SMMC7721并引起其增殖阻滯。
