目的:探討重組人IGF-1和生長抑素-14單獨(dú)及聯(lián)合應(yīng)用對甲狀腺相關(guān)眼病患者眼眶成纖維細(xì)胞增殖的影響。方法:體外培養(yǎng)甲狀腺相關(guān)眼病患者眼眶成纖維細(xì)胞,采用四甲基偶氮唑鹽(MTT)比色法檢測不同濃度及作用時間的重組人IGF-1和生長抑素-14對細(xì)胞增殖的作用。結(jié)果:50μg/L以上濃度IGF-1對眼眶成纖維細(xì)胞有促增殖作用,5nmol/L以上濃度的SST對眼眶成纖維細(xì)胞的生長有抑制作用,均呈量效和時效關(guān)系。IGF-1和SST聯(lián)合應(yīng)用時, 50nmol/L以上的SST能阻斷100μg/L的IGF-1促細(xì)胞增殖的作用。結(jié)論:IGF-1可刺激眼眶成纖維細(xì)胞增殖。SST能抑制眼眶成纖維細(xì)胞增殖,并能阻斷IGF-1促進(jìn)細(xì)胞增殖的作用,此抑制作用與濃度有關(guān),提示SST能直接通過受體后機(jī)制抑制IGF-1誘導(dǎo)的眼眶成纖維細(xì)胞增殖。
