目的 探討周期性力學(xué)刺激對體外培養(yǎng)的成肌細胞自身抗原表達的影響。 方法 取C2C12細胞根據(jù)處理方法不同分為實驗組及對照組,其中實驗組分為4個亞組,即2、4、6 d周期性拉伸組(2、4、6 d拉伸組)及1 d拉伸組。2、4、6 d拉伸組:細胞置于Flexercell 5000柔性基底加載系統(tǒng),以0.25 Hz拉伸頻率、10%拉伸幅度進行每天2 h應(yīng)力加載,連續(xù)加載2、4、6 d;1 d拉伸組:細胞置于Flexercell 5000柔性基底加載系統(tǒng),以1 Hz拉伸頻率、15%拉伸幅度拉伸1 h,23 h后收集細胞;對照組:細胞常規(guī)培養(yǎng)1、2、4、6 d。培養(yǎng)期間于倒置相差顯微鏡下觀察細胞生長情況。采用流式細胞儀檢測細胞增殖情況,Western blot檢測自身抗原DNA依賴蛋白激酶催化亞基(Ku/the catalytic subunit of DNA-dependent protein kinase,DNA-PKcs)、Mi-2(reconfigurable components deacetylase complexes of NuRD)、U1-70(A part of ATP-dependent DNA helicaseⅡ)、組氨酰-tRNA合成酶(histidyl tRNA synthetase,HRS)表達情況。 結(jié)果 經(jīng)培養(yǎng)后,1 d拉伸組可見脫落細胞,對照組1 d時無細胞脫落;2 d拉伸組細胞增殖較對照組2 d時明顯;4 d拉伸組細胞出現(xiàn)分化,6 d拉伸組細胞部分融合,與對照組4、6 d時情況一致。1 d拉伸組細胞經(jīng)單次拉伸后出現(xiàn)凋亡,相對DNA增殖指數(shù)(relative DNA proliferation index,DPI)與對照組1 d時比較,差異無統(tǒng)計學(xué)意義(t=0.346,P=0.747)。經(jīng)周期性拉伸后,2、4 d拉伸組DPI較對照組2、4 d時顯著提高(P  lt; 0.05),6 d拉伸組與對照組6 d時比較差異無統(tǒng)計學(xué)意義(t=1.191,P=0.303)。2 d拉伸組DNA-PKcs、Mi-2、HRS和U1-70蛋白相對表達量均較對照組2 d時明顯降低(P  lt; 0.05); 4 d拉伸組與對照組4 d時比較差異均無統(tǒng)計學(xué)意義(P  gt; 0.05)。4 d拉伸組各抗原蛋白相對表達量均較2 d拉伸組顯著增加(P  lt; 0.05),而對照組4 d 時均較2 d時顯著降低(P  lt; 0.05)。 結(jié)論 短期力學(xué)刺激可以抑制成肌細胞自身抗原表達,但隨時間延長,細胞分化融合以及對力學(xué)刺激的適應(yīng)使其對自身抗原表達抑制作用減弱。
