目的 探討哮喘大鼠肺臟、鼻腔、腸道黏膜免疫的相互關(guān)聯(lián)和變化規(guī)律。方法 將20只Wistar 大鼠分為對(duì)照組和哮喘組, 各10 只。采用卵白蛋白致敏和激發(fā)的方法制作哮喘模型。應(yīng)用免疫組化和原位雜交技術(shù)檢測(cè)大鼠肺組織、鼻黏膜、腸黏膜中的CD4 + 、CD8 + 淋巴細(xì)胞數(shù)量, eotaxin蛋白及其mRNA 表達(dá)。取大鼠支氣管肺泡灌洗液( BALF) 、鼻咽沖洗液、腸黏液上清液, 通過酶聯(lián)免疫吸附試驗(yàn)檢測(cè)sIgA 含量。結(jié)果 哮喘組肺組織、鼻黏膜、腸黏膜中CD4 + 和CD8 + 表達(dá)較對(duì)照組增高( P lt;0. 05) , 肺組織中eotaxin 蛋白及mRNA、鼻咽沖洗液中sIgA、腸黏膜eotaxin 蛋白表達(dá)較對(duì)照組增高( P lt;0. 05) 。其余指標(biāo)兩組間水平無顯著差異。對(duì)照組大鼠eotaxin 蛋白表達(dá)在肺組織和鼻黏膜呈負(fù)相關(guān)( r= - 0. 572, P =0. 008) , 在腸黏膜和鼻黏膜呈正相關(guān)( r =0. 638, P =0. 002) ; eotaxin mRNA表達(dá)在肺組織和鼻黏膜呈正相關(guān)( r =0. 502, P = 0. 024) , 在腸黏膜和鼻黏膜呈正相關(guān)( r =0. 594, P =0. 006) 。在哮喘組, 除了eotaxin 蛋白表達(dá)在肺組織和腸黏膜呈負(fù)相關(guān)( r = - 0. 448, P = 0. 048) , 其余指標(biāo)間無顯著相關(guān)性。結(jié)論 在正常生理狀態(tài)下, 肺、鼻、大腸之間的黏膜免疫功能可能保持密切的關(guān)系或動(dòng)態(tài)平衡。但是在哮喘發(fā)病過程中, 這種平衡被破壞。
