目的 觀察哮喘小鼠肺組織CCL1 /CCR8 mRNA 的表達(dá)及糖皮質(zhì)激素對其表達(dá)的影響, 探討CCL1 /CCR8 在哮喘發(fā)病機(jī)制中的作用。方法 30 只小鼠隨機(jī)分為對照組、哮喘組和地塞米松組, 每組10 只。哮喘組和地塞米松組用卵白蛋白致敏激發(fā)建立哮喘模型, 地塞米松組給予腹腔注射地塞米松( 2 mg/kg) , 對照組均以生理鹽水替代。測定BALF 中細(xì)胞總數(shù)和分類計(jì)數(shù), 采用酶聯(lián)免疫法檢測BALF 中IL-4 水平, RT-PCR 檢測肺組織CCR8 mRNA 和CCL1 mRNA 的表達(dá)。結(jié)果 對照組、哮喘組和地塞米松組BALF 中嗜酸粒細(xì)胞百分比[ ( 0. 6 ±0. 1) % 、( 32. 4 ±3. 4) % 和( 8.9 ±0. 9) % , P lt;0. 01] 、淋巴細(xì)胞百分比[ ( 2.9 ±0. 5) % 、( 14. 0 ±3. 0) % 和( 6. 3 ±0. 6) % , P lt;0. 01] 、IL-4濃度[ ( 5.16 ±1. 23) pg /mL、( 47. 67 ±11. 67) pg/mL 和( 19. 76 ±4. 72) pg/mL, P lt;0. 01] 差異均有統(tǒng)計(jì)學(xué)意義, 其中哮喘組較對照組顯著增高( P lt;0. 01) , 地塞米松組較哮喘組顯著降低但仍顯著高于對照組( P lt;0. 01) 。對照組、哮喘組和地塞米松組CCL1 mRNA 表達(dá)( 0.11 ±0. 06、0. 76 ±0. 16 和0. 53 ±0. 13, P lt;0. 01) 、CCR8 mRNA 表達(dá)( 0. 18 ±0. 09、1. 26 ±0. 13 和0. 79 ±0. 12, P lt; 0. 01) 差異均有統(tǒng)計(jì)學(xué)意義, 其中哮喘組較對照組顯著增高( P lt;0. 01) , 地塞米松組較哮喘組顯著降低但仍顯著高于對照組( P lt;0. 01) 。結(jié)論CCL1/CCR8 在哮喘小鼠肺組織中的基因表達(dá)增強(qiáng)。糖皮質(zhì)激素可能通過減少CCL1 /CCR8 的基因表達(dá), 減輕哮喘氣道炎癥。
