目的 探討不可分型流感嗜血桿菌(NTHi)作用于呼吸道上皮細胞株(A549),表達前炎癥細胞因子的變化及其干預措施。方法 用NTHi、NTHi+紅霉素(10 mg/L)、NTHi+慶大霉素(100 mg/L)、NTHi+地塞米松(100 μmol/L)分別感染A549細胞,用核因子κB(NF-κB)抑制劑二硫氨基甲酸吡啶(PDTC,200 μmol/L)預處理NTHi感染的A549細胞,24 h后觀察細胞的形態(tài)學改變,并檢測上清液中白細胞介素8(IL-8)、腫瘤壞死因子α(TNF-α)的水平和細胞間黏附分子1(ICAM-1)的表達。結(jié)果 NTHi作用A549細胞24 h后,引起部分上皮細胞變形、死亡。NTHi能顯著誘導A549細胞株分泌IL-8、表達ICAM-1,紅霉素能夠抑制其誘導作用;慶大霉素具有殺滅NTHi作用,但不能抑制IL-8分泌和ICAM-1表達;地塞米松能抑制A549分泌IL-8,降低ICAM-1的表達,但不能殺滅NTHi。PDTC(200 μmol/L)能阻斷A549細胞在NTHi作用后IL-8的高表達。TNF-α在各組培養(yǎng)上清中均不能檢測到。結(jié)論 NTHi顯著誘導人肺上皮細胞系表達前炎癥因子,該作用可能通過NF-κB通路。紅霉素在殺滅NTHi的同時還具有抗炎作用。
