應(yīng)用ABC免疫組化及半巢式PCR法分別對64例大腸癌組織中bcl-2的蛋白表達(dá)及重排進(jìn)行了檢測。結(jié)果:bcl-2基因異常表達(dá)與重排在大腸癌早期即已出現(xiàn),隨著病程演變,發(fā)生率顯著增加;bcl-2基因的重排在有淋巴結(jié)轉(zhuǎn)移者亦明顯多于無淋巴結(jié)轉(zhuǎn)移者。兩法聯(lián)合應(yīng)用相比較,以重排更能反映大腸癌分子特性。本實(shí)驗(yàn)結(jié)果提示:bcl-2基因參與大腸癌發(fā)生發(fā)展過程的調(diào)節(jié),并在大腸癌細(xì)胞的增殖、進(jìn)展及轉(zhuǎn)移中起重要作用;研究大腸癌bcl-2基因情況可用來預(yù)測治療效果和預(yù)后,亦為大腸癌治療設(shè)計(jì)新的抗癌藥物提供了新的靶分子。
