目的了解抗血管內(nèi)皮生長因子(VEGF)抗體對人肝癌的生長抑制作用。方法在已建立的人肝癌裸鼠皮下種植瘤的瘤灶旁,注射抗VEGF抗體,觀察腫瘤生長情況,用CD31的免疫組化方法檢測腫瘤內(nèi)部的微血管密度(iMVD),用原位末端轉(zhuǎn)移酶標(biāo)記技術(shù)檢測凋亡的腫瘤細(xì)胞。結(jié)果實驗結(jié)束后1周,實驗組和對照組腫瘤大?。ㄩL×寬)是(26.46±19.81) mm2和(105.77±17.40) mm2(P<0.001),2周后是(45.20±23.02) mm2和(150.77±77.41) mm2(P<0.05),而此時腫瘤的iMVD是(2 311±120)個/mm2和(3 900±328)個/mm2 (P<0.001); 腫瘤細(xì)胞凋亡指數(shù)是15.31% 和 6.83%(P<0.005)。結(jié)論抗VEGF抗體能通過抑制腫瘤微血管的生長,抑制實驗性人肝癌的生長,其實質(zhì)是增加了腫瘤細(xì)胞凋亡。
