目的 探討5-氟尿嘧啶(5-fluorouracil,5-FU)體外誘導(dǎo)直腸癌HR8348細(xì)胞凋亡的作用及細(xì)胞凋亡與bcl-2、bcl-xl、bax及p53表達(dá)的關(guān)系。方法 經(jīng)5-FU處理HR8348細(xì)胞24 h后,用甲基綠-派若寧染色法和TUNEL法檢測(cè)細(xì)胞凋亡情況,并用SP免疫組織化學(xué)法檢測(cè)HR8348細(xì)胞中bcl-2、bcl-xl、bax和p53的表達(dá)。結(jié)果 HR8348細(xì)胞經(jīng)5-FU作用24 h后,細(xì)胞凋亡指數(shù)(AI)明顯高于對(duì)照組,差異有顯著性意義(P<0.01)。5-FU作用不同時(shí)相bcl-2的表達(dá)評(píng)分結(jié)果與對(duì)照組比較差異均無(wú)顯著性意義; 不同時(shí)相bcl-xl的表達(dá)評(píng)分結(jié)果與對(duì)照組比較稍降低; 而bax的表達(dá)評(píng)分結(jié)果隨時(shí)間延長(zhǎng)明顯增加,24 h、36 h的表達(dá)評(píng)分結(jié)果明顯高于對(duì)照組(P<0.01),而p53在5-FU組和對(duì)照組各時(shí)相均未見表達(dá)。結(jié)論 5-FU具有誘導(dǎo)HR8348細(xì)胞凋亡的作用; 5-FU可能通過(guò)上調(diào)bax的表達(dá)并改變bax/bcl-xl的比值而誘導(dǎo)HR8348細(xì)胞凋亡。
