【摘要】目的 探討胃癌細(xì)胞外基質(zhì)中纖維連接蛋白(fibronectin, FN)的改變及其與胃癌組織微血管生成的相關(guān)性。
方法 應(yīng)用免疫組織化學(xué)EnVisonTM 法,對(duì)80例胃癌組織和20例正常胃腺體組織中FN的表達(dá)、血管內(nèi)皮生長(zhǎng)因子(vascular endothelial growth factor, VEGF)的表達(dá)及腫瘤微血管密度(microvessel density, MVD)進(jìn)行檢測(cè)。
結(jié)果 FN免疫復(fù)合物呈棕黃色分布于胃腺體基底膜及周?chē)慕Y(jié)締組織間質(zhì)中。正常組織基底膜中FN染色呈完整線性結(jié)構(gòu),結(jié)締組織間質(zhì)中FN染色呈規(guī)則的條紋狀。胃癌組織基底膜FN的完整線性染色結(jié)構(gòu)被不同程度破壞,結(jié)締組織間質(zhì)中FN染色加深,形狀不規(guī)則。胃癌組織中VEGF表達(dá)和MVD較正常組織均升高(P<0.01, P<0.05)。胃癌結(jié)締組織間質(zhì)中FN表達(dá)與VEGF表達(dá)和MVD均存在正相關(guān)性(P<0.01, P<0.05); 而胃癌組織基底膜FN線性染色缺損程度與VEGF表達(dá)和MVD均無(wú)相關(guān)性(P gt;0.05, P gt;0.05)。
結(jié)論 胃癌結(jié)締組織間質(zhì)中FN表達(dá)的增高可能與其微血管形成和發(fā)展過(guò)程關(guān)系密切,F(xiàn)N與VEGF在胃癌微血管的形成和發(fā)展的過(guò)程中可能存在相互協(xié)同和促進(jìn)關(guān)系。
