目的探討胃癌侵襲和轉(zhuǎn)移過程中黏著斑激酶(FAK)的表達(dá)與其生物學(xué)行為的關(guān)系。
方法用免疫組化SABC法檢測(cè)50例行胃癌根治術(shù)所收集的癌組織、癌旁組織、正常胃黏膜組織和胃區(qū)域淋巴結(jié)標(biāo)本中FAK的表達(dá)情況。
結(jié)果FAK表達(dá)強(qiáng)陽性率在胃癌組織中(78.0%)明顯高于正常胃黏膜組織(10.0%)及癌旁組織(20.0%),P<0.01,正常胃黏膜組織與癌旁組織中的表達(dá)差異無統(tǒng)計(jì)學(xué)意義(P gt;0.05)。FAK表達(dá)強(qiáng)陽性率在淋巴結(jié)有轉(zhuǎn)移和無轉(zhuǎn)移患者中分別是68.8%(22/32)和33.3%(6/18),前者明顯高于后者(P<0.05); 在有轉(zhuǎn)移的淋巴結(jié)中和無轉(zhuǎn)移的淋巴結(jié)中分別是87.2%(136/156)和40.9%(18/44),前者明顯高于后者(P<0.01); FAK表達(dá)強(qiáng)陽性率與胃癌浸潤深度和分化程度有關(guān),浸潤深度越深,細(xì)胞分化越低,表達(dá)強(qiáng)陽性率越高(P<0.05); 與Borrmann分型、腫瘤部位及大小無關(guān)(P gt;0.05)。
結(jié)論FAK表達(dá)與胃癌發(fā)生、發(fā)展有關(guān),F(xiàn)AK表達(dá)增高是胃癌細(xì)胞浸潤和轉(zhuǎn)移的重要原因之一,檢測(cè)癌組織中FAK表達(dá)水平,有助于進(jìn)一步了解胃癌的生物學(xué)行為,有利于治療與預(yù)后判斷。
