目的  研究RNA干擾(RNAi)對兔血管平滑肌細(xì)胞(VSMCs)bcl-2基因表達(dá)的干擾效應(yīng)。
方法  用脂質(zhì)體法將構(gòu)建的裝載靶向bcl-2基因小干擾性RNA(siRNA)的表達(dá)載體(pshRNA-bcl-2質(zhì)粒)轉(zhuǎn)染VSMCs(基因組),以轉(zhuǎn)染空載體的細(xì)胞和加DMEM的空白細(xì)胞作為對照,采用半定量RT-PCR 和Western blot法檢測bcl-2基因的表達(dá),用MTT法檢測VSMCs生長情況。
結(jié)果  轉(zhuǎn)染siRNA的表達(dá)載體可以抑制內(nèi)源性bcl-2基因在轉(zhuǎn)錄和轉(zhuǎn)譯水平上的表達(dá),基因組bcl-2的mRNA和蛋白表達(dá)較空載體組和空白對照組明顯減少(P<0.01); 基因組的VSMCs生長也較空載體組和空白對照組明顯受到抑制(P<0.01)。
結(jié)論  載體介導(dǎo)的RNAi技術(shù)可明顯抑制內(nèi)源性bcl-2基因的表達(dá)和VSMCs生長
