目的 探討丙型肝炎病毒(HCV)的F蛋白對(duì)肝星形細(xì)胞生長(zhǎng)及膠原蛋白表達(dá)的影響。方法 將含有HCV f基因的pcDNA3.1-f質(zhì)?;騪cDNA3.1空載體分別轉(zhuǎn)染肝星形細(xì)胞株LX2,G418篩選后獲得穩(wěn)定轉(zhuǎn)染的細(xì)胞,分別命名為L(zhǎng)X-f細(xì)胞或LX-p細(xì)胞,MTT法測(cè)定細(xì)胞增殖情況,ELISA法檢測(cè)細(xì)胞分泌的Ⅰ型和Ⅲ型膠原蛋白含量。結(jié)果 自培養(yǎng)24 h起,LX-f細(xì)胞各時(shí)間點(diǎn)的增殖率均大于LX-p細(xì)胞(P lt;0.01)。培養(yǎng)48 h時(shí),LX-f細(xì)胞的Ⅰ型膠原蛋白及Ⅲ型膠原蛋白分泌量分別為(25.89±0.42) ng/ml和(18.21±0.49) ng/ml,明顯高于LX-p細(xì)胞的(22.65±0.49) ng/ml和(15.29±0.62) ng/ml(P lt;0.01)。結(jié)論 HCV的F蛋白能促進(jìn)肝星形細(xì)胞增殖,并上調(diào)細(xì)胞分泌Ⅰ型和Ⅲ型膠原蛋白,誘發(fā)肝纖維化。
